Okay, I'm not usually shirty about my title, but when:

a) it's in an academic context

b) it's in response to an email that I signed "Dr."

c) it's from someone in a context where the title is definitely required

Being addressed as "Ms." ticks me off. 😛

On a related note, I was recently in an IEP meeting for my kiddo and the school psychologist was like "you're Ms. Papke?" and I was like "Dr. Papke if we're doing titles, but you can call me Julia". Like, please, call me by my first name but if we're doing titles then use the right one.
